Difference between 1:1 copy and a program that allows you to play backup copy?

If I have made a 1:1 copy of a game, do I still need to have a program like daemon tools to play that game? If so, why?

NO, if you have a 1:1 copy, this means you have a cd that is 100% identical to the original, so you don't need anything.
You can "verify" if you have a working (and probably 1:1 copy then) copy by using daemon-tools to mount the IMAGE-FILE.
