I don't know about Puzzle Bobble but I got Wipeout pure to run off the MS with Twisted metal Head on in the drive....
Do you have any original Game UMD in the drive when you are trying to boot Puzzle Bobble?? I believe that Lumines is the only game you can run off the MS without a substitute game in the UMD Slot. Edit: Wipeout Pure does hang at the loading screen if there is no UMD in the drive....maybe this is your problem too.
