Nope, SegaSmash plays USA roms only. Launch Tool forces a "U" inside JAP roms, so they can be played. The only games that need to be patched are PAL, because they have not only a different protection for each game, but they also run at 50Hz.
I am 100% sure of this. I have built my SegaGen collection when it was released more than 2 years ago and I needed to set a U in JAP roms, and patch every PAL rom with its own IPS. Only a very few non PAL games are protected beyond country check. 700+ unique games manually tested.
